Background
GSPT1, also known as eukaryotic release factor 3a (eRF3a), is a GTP-binding protein that plays a crucial role in both translation termination and cell cycle regulation. It functions by binding with eRF1 to dissociate proteins from the ribosome after translation, thus regulating cell cycle progression from the G1 to S phase. GSPT1 is overexpressed in various cancers, including gastric, colorectal, lung, and breast cancers, making it a promising target for cancer therapy. Additionally, GSPT1 is involved in cytoskeleton organization and apoptosis. Its degradation through molecular glue degraders, such as CC-885, has been shown to exert strong antitumor activity.
